摘     要


應用小鼠進行 粉刺配方 急性毒性實驗表明： 小鼠口服灌胃 粉刺配方 在  186g生藥 /kg 劑量下，連續觀察 7 天， 無一動物死亡。小鼠全身狀況、飲 食、攝水和體重增長均正常。動物處死屍解無病理學改變。


LD50* > 186g 生藥/kg。

按體重計，小鼠口服灌胃 粉刺配方 最大耐受量為臨床成人用的 651 倍。提 示該藥急性毒性低，臨床用藥安全。


試驗資料

(一) 實驗材料：

1.試驗藥物： 粉刺配方 （由土茯苓 12%、連翹 7%、金銀花 7%、牡丹皮 6%、蒲公英 7%、天花粉 7%、海藻 7%、浙貝母 4%、丹參 10%、當歸 4%、山楂 7%、陳皮 4%、桃仁 4%、薏苡仁 14% 組成），實驗時煎煮並 濃縮成 4.65g生藥/ml 供實驗用。

2.實驗動物： NIH 小白鼠，體重 19∼22g，雌雄各半，由廣東省醫學實驗 動物中心提供。動物合格證號：99A030。符合壹級標準，正常飼養 3 天 後供試。


(二) 實驗方法與結果：

1.給藥途徑： 採用小鼠口服灌胃給藥。

2.劑量設計：經預試， 用最高濃度和容積的 粉刺配方 液灌胃量，給予小 鼠灌胃，小鼠無一死亡， 粉刺配方 急性毒性低， 未能測得 LD50，故本 品僅進行最大耐受量試驗。

3.實驗方法：取 NIH 小白鼠 20 只，體重 19∼22g，雌雄各半，給予上述  粉刺配方 液灌胃，灌胃容積為 0.4ml/10g，1 次/天，給藥後連續觀察  7 天，並記錄小鼠的毒性反應及死亡數。

4.實驗結果：結果表明：實驗小鼠連續觀察 7 天，無一動物死亡，其全身 狀況、飲食、攝水和體重增長均正常，動物處死屍解無病理學改變。

小鼠口服 粉刺配方 LD50 > 186g生藥/kg。

若 粉刺配方 成人(平均體重以 70kg 計)臨床用量為 20g/天，按體重計：小鼠 (平均體重以 20g 計) 口服灌胃 粉刺配方 的最大耐受量則為臨床成人用量的 651倍**， 見表。

(三) 結論：
